These improvements greatly increased both the product yield and quality with CHO cell line products having seen nearly a 100 fold increase in titers compared to similar processes developed in the 1980’s (Lai et al., 2013). Despite these past advancements, there is still room for improvement for cell line selection in a number of areas. One of the major challenges for cell line development is lengthy timelines, with the average timeline for cell line development being 6 to 12 months (Lai et al, 2013). Clone screening technologies may be able to significantly reduce cell line development, with recent advancements helping increase throughput and more targeted selection through automation. Berkeley Lights has created a cell selection instrument, the Beacon system, that allows for automated end to end cell line development using optofluidics. The instrument is capable of cell culture, assaying, and cell selection, allowing for isolation of a desirable high producing clone in a much shorter period of time compared to more traditional methods. The Beacon system was used for cell line development for antibody production for SARS-CoV2-2 in a matter of 25 days, that were used for AstraZeneca preventative treatments for Covid-19 (Zost et al., 2020). The Beacon System has provided very promising results so far. However, most of the Beacon applications have been for CHO cell lines.
